Jermaine Eluemunor was born on December 13, 1994, in Chalk Farm, London, to a Nigerian father and English mother. He attended Haverstock School in London and grew up playing rugby and cricket. His interest in American football began after watching a 2007 NFL International Series game between the Miami Dolphins and New York Giants at Wembley Stadium in London.
At age 14, Eluemunor moved to the United States with his father and began playing American football at Morris Knolls High School in Denville Township, New Jersey. There, he played on both the defensive and offensive lines and also competed as a wrestler, achieving a 33–4 record as a senior.

Despite not being recruited by college football programs out of high school, Eluemunor attended Lackawanna College in Pennsylvania from 2012 to 2013, where he earned all-conference second-team honors in 2013. After attracting recruitment interest from 35 college programs, he initially committed to UCLA, then Arkansas, before ultimately enrolling at Texas A&M University.
At Texas A&M, Eluemunor redshirted in 2014 and served as a backup right guard in 2015, making his first career start during the Music City Bowl that year.
