Hamler redshirted as a freshman at Penn State in 2017 due to the ACL injury he sustained while in high school. He returned to the field in 2018 and made his collegiate debut against Appalachian State. In that game, he returned a kickoff 52 yards with less than two minutes remaining to help spark Penn State’s game-tying drive and capped it off with a 15-yard touchdown reception with under a minute left, finishing with three catches for 67 yards and a touchdown.

Against Ohio State during his freshman season, Hamler earned Big Ten Conference Freshman of the Week honors after recording four receptions for 138 yards, including a 93-yard touchdown. Through five games in 2018, he recorded 13 receptions for 308 yards and four touchdowns. His first career touchdown came in Week 8 against the Los Angeles Chargers when he caught a pass with no time remaining on the clock to help the Broncos win 31–30.
